Mechanism of Action

Multi-pathway metabolic activity

Berberine acts through several converging pathways rather than a single receptor target. It activates AMP-activated protein kinase (AMPK) in liver, skeletal muscle, and adipose tissue, improving cellular glucose uptake and lipid handling. In the liver, berberine upregulates LDL receptor (LDLR) expression via AMPK-ERK signaling and inhibition of miR-27a, improving LDL cholesterol clearance. Berberine also interacts with gut microbiota, promoting short-chain fatty acid (SCFA) production — including butyrate — that contributes to its systemic metabolic effects, a relevant consideration given berberine's inherently low oral bioavailability.
